From Manila: fly to the port city of Calapan (Mindoro), then drive by car or van roughly 2-3 hours to the Naujan area on the eastern side of the island (distances and times vary by ferry schedules). From Calapan, take the national highway toward Naujan and Victoria; the lake lies a short local drive from those town centers. Local tricycles or hired driver-guides are useful for the final leg.
